If memory serves, Ryuko needs to be pretty thoroughly destroyed to get rid of her, right? Esdeath could combo her time stop into a more devastating blow, but every time we see her use it it's to set up a simple brain stab. If she landed a good hit before using the time stop and understand that she needed a big hit to win, that might allow her to win here, but I don't remember her usually using attacks that freeze opponents solid and destroy them at a level that would kill Ryuko. I might be misremembering. In any case, Ryuko is faster, but only a tier, so it's not out of the realm of possibility for Esdeath to land a hit. Right now leaning towards Ryuko, but I'm open to discussion

Yeah, she's absurdly tough and with a strong healing factor. She's been cut in half, had her heart ripped out of her, she's punched into her own skull and ripped a mind control tool right off her brain, torn off an outfit that had been more or less fused with her skin and thus took large parts of her own skin off, and has been pierced through with dozens of stabs from large weapons that tore holes all over her body but she still endured and healed from all of those pretty much instantly. So yeah, it would take a massive attack that like vaporized her utterly to do it, I feel.

And that’s assuming Esdeath would be able to survive Ryuko’s attacks in turn. And she can casually slice through two skyscrapers at once.
